چکیده

This study aimed to explore the effect of treadmill exercise on anxiety in rats. Thirty SPF male rats aged 2 months with a body mass (225±25) g were randomly divided into control group (CG, n=10), chronic sleep deprivation (CSD, n=10) and (CSD+E, after adaptive feeding for 1 week. The CSD model CSD+E was made by multi platform water environment method. Sleep 18 h per day (from 12:00 pm. 6:00 am. next day) 8 weeks. running anxiety-like behaviour examined open field test (OFT) elevated plus maze (EPM) experiment. Hematoxylin-eosin (HE) staining, Annexin V/PI flow cytometry, immunofluorescence Western blot, RT-qPCR other methods used detect effects morphology hippocampus, apoptosis related factors caspase-12, Bax, Bcl-1, inflammatory (IL-6, TNF-a), Wnt β- catenin, p- β-catenin. results HE staining showed that brain tissue structurally intact, thick layers cone cells, relatively dense, neatly arranged compact, cell edge structures intact clearly visible, no obvious abnormal changes. layer thin, low density, disorganized sparse arrangement, blurred edges. symptoms above pathological changes gradually alleviated. OFT compared CG group, number activities central region significantly reduced (P<0.01), total distance shortened (P<0.01). EPM OT CE decreased CCK-8 activity neurons CA1 area hippocampus while increased (P<0.01); cytometry (P<0.01);Western blot IL-6, IL-1β TNF-a highly expressed Bcl-1 lowly hippocampal tissues Wnt, β-catenin p-β-catenin Gsk-3βprotein expression higher (p<0.01). caspase-12 mRNA, Bax IL-6 mRNA Compared Gsk-3βmRNA Our findings indicated weeks aerobic improved depression increasing neuronal activity, inhibiting apoptosis, reducing response activating Wnt/β-catenin pathway.

A clinical genetic method to identify mechanisms by which pain causes depression and anxiety

BACKGROUND Pain patients are often depressed and anxious, and benefit less from psychotropic drugs than pain-free patients. We hypothesize that this partial resistance is due to the unique neurochemical contribution to mood by afferent pain projections through the spino-parabrachial-hypothalamic-amygdalar systems and their projections to other mood-mediating systems. New psychotropic drugs for ...

Modeling the Regulatory Mechanisms by Which NLRX1 Modulates Innate Immune Responses to Helicobacter pylori Infection

Helicobacter pylori colonizes half of the world's population as the dominant member of the gastric microbiota resulting in a lifelong chronic infection. Host responses toward the bacterium can result in asymptomatic, pathogenic or even favorable health outcomes; however, mechanisms underlying the dual role of H. pylori as a commensal versus pathogenic organism are not well characterized. Recent...

Resveratrol Ameliorates the Anxiety- and Depression-Like Behavior of Subclinical Hypothyroidism Rat: Possible Involvement of the HPT Axis, HPA Axis, and Wnt/β-Catenin Pathway

Metabolic disease subclinical hypothyroidism (SCH) is closely associated with depression-like behavior both in human and animal studies, and our previous studies have identified the antidepressant effect of resveratrol (RES) in stressed rat model. The aim of this study was to investigate whether RES would manifest an antidepressant effect in SCH rat model and explore the possible mechanism. A S...

Oestrogen and parathyroid hormone alleviate lumbar intervertebral disc degeneration in ovariectomized rats and enhance Wnt/β-catenin pathway activity

To investigate the mitigation effect and mechanism of oestrogen and PTH on disc degeneration in rats after ovariectomy, as well as on Wnt/β-catenin pathway activity, thirty 3-month-old rats were ovariectomized and divided into three groups. Ten additional rats were used as controls. Eight weeks later, the rats were administered oestrogen or PTH for 12 weeks, and then discs were collected for te...

Treating Depression and Depression-Like Behavior with Physical Activity: An Immune Perspective

The increasing burden of major depressive disorder makes the search for an extended understanding of etiology, and for the development of additional treatments highly significant. Biological factors may be useful biomarkers for treatment with physical activity (PA), and neurobiological effects of PA may herald new therapeutic development in the future. This paper provides a thorough and up-to-d...
